Experimental Autoimmune Uveitis: An Intraocular Inflammatory Mouse Model

چکیده
Experimental Autoimmune Uveitis (EAU) is driven by immune cells responding to self-antigens. Many features of this non-infectious, intraocular inflammatory disease model recapitulate the clinical phenotype posterior uveitis affecting humans. EAU has been used reliably study efficacy novel therapeutics, their mode action and further investigate mechanisms that underpin progression disorders. Here, we provide a detailed protocol on induction in C57BL/6J mouse - most widely organism with susceptibility disease. Clinical assessment severity will be demonstrated using fundoscopy, histological examination fluorescein angiography. The procedure involves subcutaneous injection an emulsion containing peptide (IRBP1-20) from ocular protein interphotoreceptor retinoid binding (also known as retinol 3), Complete Freund's Adjuvant (CFA) supplemented killed Mycobacterium tuberculosis. Injection viscous back neck followed single intraperitoneal Bordetella pertussis toxin. At onset symptoms (day 12-14) under general anesthesia, fundoscopic images are taken assess through examination. These data can directly compared those at later timepoints peak 20-22) differences analyzed. same time, allows investigator potential vessel permeability damage induced other strains both wildtype or genetically modified combined therapies offering flexibility for studying drug and/or mechanisms.

Mouse models of experimental autoimmune uveitis.
The mouse model of experimental autoimmune uveitis, induced by immunization of mice with the retinal protein IRBP, was developed in our laboratory 20 years ago and published in 1988. Since that time it has been adopted by many investigators and has given rise to many studies that helped elucidate genetic influences, dissect the basic mechanisms of pathogenesis and test novel immunotherapeutic p...
متن کاملAnti-inflammatory role of IL-17 in experimental autoimmune uveitis.
Previous studies have shown that IL-17 is a strong proinflammatory cytokine and that IL-17-producing autoreactive T cells play a major role in the pathogenesis of autoimmune diseases. In a previous study, we showed that injection of experimental autoimmune uveitis-susceptible mice with anti-IL-17 Abs blocked subsequent disease development. To determine whether administration of IL-17 to experim...
متن کاملExperimental mouse model of tuberculous uveitis
Results The i.v. and i.n. challenged mice showed significant DTH response. The CFU enumeration in eye did not show any bacilli initially by 30th day however tubercle bacilli could be recovered from the eyes of i.v. infected mice by 45 day at the time of death. Histopathology of these eyes showed inflammation with cluster of lymphocytes. The RTPCR for IS6110 gene from the eyes of i.n. infected a...
متن کاملPathogenesis of innate immunity and adaptive immunity in the mouse model of experimental autoimmune uveitis.
Experimental autoimmune uveitis, a well-established model for human uveitis, is similar to human uveitis in many pathological features. Studies concerning the mechanisms of experimental autoimmune uveitis would cast a light on the pathogenesis of human uveitis as well as the search for more effective therapeutic agents. The cellular components of innate immunity include natural killer cells, ga...
متن کاملIntraocular antibody synthesis during experimental uveitis.
Determination of intraocular antibody synthesis against certain microorganisms is a diagnostic aid in identifying the causative agent in clinical uveitis. Little is known, however, concerning the kinetics and specificity of antibodies produced during intraocular inflammation. To investigate this subject we induced uveitis in rabbits by injecting small amounts of human serum albumin (HSA) into t...
